Agar of Asian origin has been consumed for centuries as a basic element in the traditional diets of numerous populations in the Far East.
The different types of algae from which agar can be obtained (Gelidium, Gracilaria, Pterocladia and Gelidiella) may be used to create products with different characteristics, although the most common genera for this purpose are the Gelidium and Gracilaria.
The broad array of food agar allows its use in a wide number of formulations. Agarose
Agarose is a polysaccharide formed by the union of β-D-galactopyranose and 3,6-anhydro-α-L-galactopyranose linked by 1-4 bonds. This basic repeating unit, called agarobiose, forms long chains. This structure contains charged groups, such as sulfate and pyruvate, and uncharged groups, all of which are responsible for many of the properties of agarose. These properties can be adjusted to meet specific needs through the careful selection of raw materials.
Some important properties of agarose are:
- Gel strength: Force necessary to break a gel, expressed in g/cm2 .
- Gelling temperature: Temperature at which an agarose solution forms a rigid gel.
- Melt temperature: Temperature at which the gel becomes a solution.
- EEO (electroendosmosis): Measure of the number of charged groups present in the agarose.
Standard agarose is ideal for the rapid routine separation of DNA and RNA fragments, as well as for PCR products, plasmid preparation and for detection, cloning and transfer techniques.

Low melting point (LM) agaroses are obtained by organic synthesis. The main properties of these agaroses are their low melting and gelling temperatures compared to standard agaroses. The low melting temperature allows the recovery of intact and biologically active nucleic acids, not damaged by the temperature below the denaturation temperature. The low gelling temperature ensures that the agarose will be in a liquid state in a temperature range in which in-gel manipulations can be performed without prior DNA extraction.
Characteristics:
- Less gel strength than standard agaroses. Gels can nevertheless be easily handled.
- Lower gelling/melting temperatures than standard agarose.
- Greater clarity (gel transparency) than gels prepared with standard agarose.
